Most drugs are more or less bound to proteins. Basic drugs tend to bind the acidic alpha-1 acid glycoprotein (AAG), while acidic and neutral drugs tend to bind albumin, which is basic. Extent of plasma protein binding may significantly influence the distribution, clearance, and efficacy of a drug.

Our plasma protein binding assay is based on equilibrium dialysis.

After incubation, both the plasma and buffer samples are analyzed by LC-MS/MS for drug concentration. The percent protein binding of a drug is then calculated:

% Protein Binding = 100 * (Cplasma - Cbuffer) / Cplasma

Warfarin and propranolol are used as controls in our protein binding assay.
